Pupas de Chrysomya megacephala (Fabricius, 1794) (Diptera, Calliphoridae) crioconservadas em soluções de glicerol e dimetil sulfóxido como substrato de criação de Nasonia vitripennis (Walker, 1836) (Hymenoptera, Pteromalidae)

Abstract

Substances glycerol and dimethyl sulfoxide (DMSO) were evaluated as cryoprotective in the storage of the Chrysomya megacephala (Fabricius, 1794) (Diptera: Calliphoridae) pupae at low temperatures. It were investigated five concentrations (0.1, 1, 2, 3 and 4 mol.) at -20 oC (freezer) and –196 oC (fase liquid of the liquid nitrogen- LN). Control groups were constituted by non-frozen pupae (fresh) and pupae freezing only with phosfate buffer saline (PBS). Information was obtained about the reproductive performance of Nasonia vitripennis
(Walker, 1836) matrices reared on these pupae. These analyses revealed rates of parasitoidism efficiency lower than 3 % at glycerol NL (3 and 4 mol) and DMSO Freezer (0.1, 2, 3 and 4 mol). There were not emergence in the frozen pupae belong to DMSO NL and glycerol NL – PBS; 0.1, 1 and 2 mol. The freezing with glycerol at – 20 oC showed better rates of parasitoidism efficiency than others groups. Methods used in this work with cryoprotectives
glycerol and DMSO were ineffective in the cryopreservation of C. megacephala pupae when compared with others works. The influency of biotics and non-biotics facts were discussed.
